Abstract

背景：

慢性炎症通过促进免疫抑制性肿瘤微环境(TME)驱动肿瘤进展。磷脂酶A2（PLA2）通过产生生物活性脂质介质来促进这一过程。本研究探讨了具有抗肿瘤潜力的临床PLA2抑制剂异甘草酸镁（MgIG）在重塑胃癌炎性TME中的疗效和机制。

方法：

在小鼠中建立鼠胃癌(MFC)同基因同种异体移植物以评估MgIG的抗肿瘤作用。在MFC和人胃腺癌(AGS)细胞系中检测细胞活力。通过定量实时聚合酶链反应（qRT-PCR）定量肿瘤中的细胞因子和趋化因子表达。通过免疫组织化学(IHC)评估TME内的关键免疫细胞浸润。Western blotting用于研究MgIG对信号通路的下游影响。使用来自MgIG处理的肿瘤细胞的条件培养基（CM）培养鼠巨噬细胞（RAW264.7）和人单核细胞白血病（THP-1）细胞系，并通过蛋白质印迹、免疫荧光（IF）和酶联免疫吸附测定（ELISA）分析极化标志物。磷脂酶A2 IVA组过表达（Pla2g4a在MFC细胞和PLA2G4A在AGS细胞中）以验证机制途径。使用学生的t方差检验或分析（ANOVA）。生存差异通过对数秩检验评估。

结论：

MgIG通过重塑炎性TME来抑制胃癌进展，从而增强抗肿瘤免疫并抑制免疫逃避。MgIG增强细胞毒性T细胞浸润，并关键地逆转巨噬细胞从M2样表型极化至M1样表型，潜在地通过抑制PLA2-NF-κ B信号传导轴，以及相关的TLR-MYD88途径的抑制。这些发现表明MgIG具有免疫调节功能，并表明其作为抑制炎症驱动的胃癌的治疗策略的潜力，值得进一步的临床研究。
